
Apoptosis, also known as programmed cell death, is a highly regulated cellular process that plays roles many areas such as development, stress response, and cancer. It involves the activities of many protein players. In the intrinsic pathway, Bcl-2 family members promote or prevent mitochondrial permeability. IAP proteins are deactivated, cytochrome c is released and binds with apaf-1 to form the apoptosome. Apoptotic caspases are then activated beginning with caspase 9. In the extrinsic pathway, TNF alpha, Fas ligand and other cytokines mediate signaling with TNF family receptors and death domain proteins to activate caspase 8.
