DNA synthesis-based approaches utilize nucleoside analogs like BrdU or EdU, which are incorporated into newly synthesized DNA strands, marking cells actively undergoing replication. Proliferation can also be evaluated by detecting the expression of specific proteins associated with cell cycle progression, such as Ki-67 and PCNA. Additionally, metabolic assays offer an indirect assessment by quantifying cellular metabolic activity, commonly through the enzymatic reduction of substrates like resazurin or tetrazolium compounds, or by measuring intracellular ATP levels as a proxy for cell viability and growth.
