Detection

Absorbance by spectrophotometer

Results Summary

Using this reagent allowed me to determine the amount of viable tumor cells remained following co-culture with my T cells. The reagent was able to produce signal that made clear the amount of live tumor cells were in my samples. This took some optimization as I realized the absorbance wavelength of formazan overlaps with phenol red, so I had to use phenol red free media during the incubation with the reagent.

Summary

The Good

Very simple workflow.

The Bad

Very sensitive so replicates are important as well as good pipetting technique.

The Bottom Line

This reagent allows a very simple workflow for evaluating cytotoxicity. With optimization, I am able to use this reagent for my needs.
