To assess the cytotoxicity of inhibitors and antibiotics in primary human cells, I utilized a cell proliferation assay kit. This kit simplifies the process of working with a 96-well plate by reducing the required reagent volume and time. At the end of the incubation period, the plate is read using a plate reader. It is a highly sensitive kit and is also useful for conducting time-dependent tests.

Washing steps are not required to perform this assay.
Cells treated with high concentration (1 ug/ml) of puromycin antibiotics were observed with cytotoxicity compared with cells without any treatment.

Assay doesn't require washing of cells before adding the reagent.
It's a bit expensive.
For consistent results, I always prefer to use this proliferation kit in my experiments.
