A type of solid-phase extraction, the principle relies on nucleic acid binding to a solid surface, such as silica gel. Once the cellular lysate is eluted, addition of buffers with varying conditions, such as in pH or salt concentration, facilitate nucleic acid binding and removal of unwanted particles. Spin-based kits that utilize 96-well plates for centrifugation are also available for DNA extraction workflows that require higher throughputs.
