Company:

New England Biolabs

Product Name:

Monarch Gel Extraction Kit

Catalog Number:

T0120S

I used New England Biolabs' Monarch Gel Extraction kit to excise DNA from the 1% agarose gel and then purify it. Yield was good with 80 % recovery for 8 kb plasmid construct with 260:280 ratio of 1.88. But 260:230 was very poor and always getting around 0.68 which gives indication of still gel contamination.

Experimental Design and Results Summary

Application

Isolation and purification of DNA from agarose gels

Starting Material

400 mg of agarose gel slice containing expected DNA bands per reaction

Tips

None

Results Summary

Yiled was very good and can elute in very low volume (20 microliter) as compared other conventional kits. Recovery was 80% for my 8 kb plasmid DNA. Howere quality was not great especially for 260:230 ratio was poor (around 0.60 to 0.70).

Summary

The Good

Easy to follow protocol, affordable price, less hands-on time, recovery was good and can elute in low volumes

The Bad

Only bad thing is quality for 260:230 ratio was poorer and indicates still some gel contamination remaining.

The Bottom Line

It is just OK to have in the lab if you are not worried too much about the quality of DNA.
