This kit was used to quantify SA-β-gal levels in proliferative and irradiated human brain microvascular endothelial cells (HBMECs), according to manufacturer's instructions. We also used in different vascular cells (HUAECs, bENd.3 cells).

While leaving the cells at 37°C in a dry incubator (No CO2), add parafilm to avoid evaporation.

Results Summary

SA-β-gal levels in proliferative and irradiated cells were assessed by colorimetric X-gal using the Senescence β-Galactosidase Activity Assay Kit. Cells were counterstained with DAPI (2 ng/μL) for nuclei detection. 10–15 random fields were captured at 5× magnification using Leica DM5500 light microscope. Images were quantified blindly by manual counting the number of SA-β-Gal positive cells normalized to the total number of nuclei.
