I use GAPDH antibody as loading control for total cellular protein fractions or soluble protein fractions regularly under various stress conditions.
Western blot
Mouse, Human cell lines
1:10000 in 1% BSA, Overnight at 4°C
5% BSA in Tris-buffered Saline Tween-20
1:10000 HRP-conjugated anti-Mouse IgG, 1 hour at RT
ECL detection in documentation system (Vilber Lourmat).
The level of GAPDH does not change under various treatments and acts as good loading control for protein gels. GAPDH is a soluble protein and so works great with total and soluble protein fractions and is not detected in insoluble protein fractions.
