We extract RNA from large numbers of mouse retina samples and need to use reverse-transcription PCR to make cDNA from this RNA. The cDNA is then run on qPCR to detect the expression of our target genes.

Protocol Overview

RNA is first treated with DNAse and incubated at 37 degrees celcius for 20 minutes. Then a stopping buffer is added and the samples are spin for 2 minutes. The liquid is removed and added to the master mix made from the kit reagents (kit has recipe). Samples are run on thermo cycler according to the reaction protocol. Samples are then treated to RNAse H (1uL into 20uL of the sample) and incubated at 37 degrees celcius for 30 minutes.

Tips

Make all RNA into cDNA instead of splitting the same so there is no variability.

Results Summary

We made clean and stable cDNA from our RNA samples quickly and efficiently. The samples will not contain any RNA or genomic DNA contaminants.
