Real-Time PCR Thermal Cyclers/ Thermocyclers, also referred to as Quantitative PCR (qPCR) calculate PCR amplification as it occurs. Using smaller amounts of starting material and combining nucleic acid amplification and detection allows for efficiency and eliminates the post-amplification process. Real-time PCR thermocyclers can be used with both DNA and RNA. When using RNA, reverse transcriptase is used to quantify the amount of RNA in the starting sample. Relevant applications include quantitative gene expression analysis, SNP analysis, drug target validation and genotyping. Many laboratories are turning to real-time PCR thermal cyclers and some important features to consider are compact size, individually programmed wells, heated lids to prevent condensation, higher throughput and software integration. LEDs are often the excitation source and detection is provided via fluorescent DNA binding dyes or probes. Most real-time PCR thermocyclers boast an average 2-hour run time and a “fast” 40-minute run time. . Be sure to inquire what block format or well capacity a real-time PCR thermal cyclers features. Some may offer multiple configurations, such as 2 x 48 wells, 96 wells or 384 wells.
