Assay Principle

Poly (ADP-ribose) polymerases (PARPs) catalyze the transfer of ADP-ribose units from NAD+ onto target proteins, most notably histones, in response to DNA damage, making this enzyme family central to DNA repair signaling and genome stability. A PARP assay typically uses histone-coated microplate wells as the substrate; active PARP enzyme transfers biotinylated or otherwise labeled ADP-ribose units from a modified NAD+ substrate onto the histone-coated surface, and the resulting poly(ADP-ribosyl)ation is detected using a streptavidin-conjugated reporter, producing either a colorimetric or chemiluminescent signal proportional to enzyme activity. Because the assay directly links substrate modification to a labeled NAD+ analog, it provides a straightforward readout for both measuring baseline PARP activity in cell or tissue extracts and screening small-molecule PARP inhibitors, where reduced signal in the presence of a test compound indicates inhibitory potency.

Colorimetric formats generally offer a lower per-plate cost and simpler instrumentation requirements, while chemiluminescent formats provide greater sensitivity and dynamic range, an advantage when working with low PARP activity samples or when screening for weak inhibitors. Because the histone-coated well format relies on a pre-immobilized substrate, plate-to-plate and lot-to-lot consistency in histone coating quality is a meaningful source of assay variability, so including both a positive control (active PARP enzyme) and a known inhibitor control on each plate is standard practice. Some isoform-specific kits (targeting PARP family members beyond PARP1, such as tankylases) use distinct substrate and detection chemistries tailored to that isoform's particular ADP-ribosylation target, which may differ meaningfully from the classic histone-based PARP1 format.

The primary choice when selecting a PARP assay kit is colorimetric versus chemiluminescent detection, and this should be driven by required sensitivity and existing plate reader capability rather than cost alone, since chemiluminescent kits generally justify their higher per-test cost in low-activity or high-throughput screening contexts. For PARP inhibitor drug discovery programs, confirming that a kit's assay window and Z-factor have been validated for high-throughput screening formats is an important practical consideration before committing to a large compound library screen. Researchers working on non-PARP1 family members should confirm that a kit is specifically validated for that isoform's substrate specificity, since much of the commercially available PARP activity kit landscape is built around the well-characterized PARP1/histone system rather than other family members. As with any activity assay, it's good practice to review the full protocol before deciding on a kit, since the manual often reveals practical details, such as incubation times, control requirements, and data analysis steps, that aren't captured in the product summary alone.
