Endonuclease I may refer to the enzymes T7 Endonuclease I or Flap endonuclease 1. T7 endonuclease I catalyzes the cleavage of DNA that is not perfectly matched. However, it does not recognize all mismatches and generally works best for cytosine mismatches. It can also cleave cruciform DNA structures, Holliday structures or junctions, heteroduplex DNA, and nicked double-stranded DNA. Cleavage occurs at the first, second or third phosphodiester bond that is 5´ to the mismatch. Flap endonuclease 1 is involved in DNA replication and repair. Encoded by the FEN1 gene, it is a structure-specific nuclease with 5'-flap endonuclease and 5'-3' exonuclease activities. It can remove 5’overhangs (flaps) and process the 5’ ends of Okazaki fragments in the lagging strand. Substrate specificity allows FEN1 to process intermediates of Okazaki fragment maturation, long-patch base excision repair, telomere maintenance, and stalled replication fork rescue.
