In Situ Hybridization Blockers In situ hybridization (ISH) blockers are reagents for minimizing non-specific binding and unwanted background signals. Examples include DNA from calf thymus, salmon sperm, and herring sperm, which are used in pre-hybridization and hybridization solutions. These DNA solutions block the non-specific binding of ISH probes to surfaces. Endogenous peroxidases and phosphatases may interfere with the enzyme-substrate-based detection methods, such as HRP and alkaline phosphatase.
