cDNA synthesis, also known as reverse transcription, generates DNA from an RNA template. Unsatisfactory cDNA yields can arise from low RNA concentration, target gene complexity, and reagent inconsistencies. cDNA synthesis kits resolve these issues with reagent uniformity, random primer supplements, and quality tested positive controls. The only required component is the RNA template, which is added to the prepared kit reagents, pre-mixed oligos, and reverse transcriptase. Two-step reverse transcription kits are available for total RNA, polyadenylated tail RNA samples, and microRNA. Custom primers can be used in place of the kit-supplied random primers, allowing researchers to manipulate cDNA strand synthesis for maximum yield and accuracy. When choosing a cDNA synthesis kit, keep in mind the size of the transcribed fragment, compatibility with downstream applications, and number of reactions per kit. Some kits yield first-strand cDNA of 200bp (base pairs), while others are capable of generating more than 10,000 bp of final product.
