Our lab uses CD26 in our T cell panel. It can be very useful in conditions where CD26 is lost, such as Sezary Syndrome / mycosis fungoides. Using CD26 with PE was needed to ensure that we get a good, strong signal on positive T cells.

Experimental Design and Results Summary

Applications

Flow cytometry

Sample

Blood and bone marrow

Primary Incubation

10-20 minutes @4°C

Blocking Agent

2% FBS

Secondary Incubation

Tertiary Incubation

Detection

Blue (488nm) laser-equipped cytometer

Results Summary

Although the results on T cells were ok (as shown in the dot-plot, and on the single-parameter histogram - on which only the lymphocyte gate is displayed), we noticed a positive dim signal on monocytes. We did not expect this, and we are not sure why it happened.

Summary

The Good

Was positive on cells of interest.

The Bad

Maybe non-specifically binging to monocytes.

The Bottom Line

This is a good antibody, but its use will depend on the lab's context of use. We think it was satisfactory for our own applications.
