Company:

BD Biosciences

Product Name:

BD Pharmingen™ APC Mouse Anti-Human CD13

Catalog Number:

557454

Clone Number:

WM15

Our lab uses CD13 as one of its core myeloid markers. Previously, we used CD13 PE (from the same vendor) and were happy with the results. We had to switch to CD13 APC. Unfortunately, it did not perform in the same way.

Experimental Design and Results Summary

Applications

Flow cytometry

Sample

Blood and bone marrow

Primary Incubation

10-20 minutes @4°C

Blocking Agent

2% FBS

Secondary Incubation

None

Tertiary Incubation

None

Detection

Red (633nm) laser-equipped cytometer

Results Summary

The monocytes and granulocytes (both of which are typically positive for CD13) appeared positive, but with a much lower stain index and signal to noise ratio compared to what we are used to with CD13 PE. The antibody works (as shown in the plot) but it is not easy to resolve the dim positive populations.

Summary

The Good

Easy to titrate.

The Bad

Relatively poor resolution.

The Bottom Line

The best advice I can give is to try a different antibody/clone if the signal you are getting is dim.
