Flow Cytometry
Mouse LN, Spleen
Block: 1:50 in 100uL at 37C for 30 min
Fc Block (aCD16/32)
L/D: Zombie NIR 1:4000 in 100 uL at RT for 20 min
Fix perm: FoxP3 fix/perm kit at 4C for 30 min; IC stain: 1:500 in 100uL perm wash buffer at RT for 1hr
Spectral flow cytometry (Cytek Aurora)
This is a relatively good antibody for identification of T follicular helper cells in lymphoid tissues using spectral flow cytometry. Not the brightest or cleanest analysis using conventional flow cytometers but works great on the Cytek Aurora.
