Our assay focuses on the detection of CD27 on normal cells and how it changes in malignancy. We were also interested in testing CD27 on plasma cells as a marker for plasma cell dyscrasia and multiple myeloma.

Applications

Flow Cytometry

Sample

Peripheral blood (human)

Primary Incubation

10-15 minutes @ Room Temp

Blocking Agent

1% FBS

Flow cytometer equipped with violet laser

Results Summary

As shown in the dot-plot, the antibody produced reasonable, albeit dim, staining on the lymphocyte population (in red). Presumably, these are T cells. However, the dim signal means that it's very hard to distinguish dim populations, and hence difficult to appreciate the subtle changes seen in leukemia and lymphoma.

Summary

The Good

Easy to optimize. ASR.

The Bad

Expensive. Dim signal.

The Bottom Line

This antibody may work for many common applications, and labs should certainly try it when they need CD27 in a violet-laser channel.
