Easy-to-use fluorescent probe, binding to mitochondrial superoxide in live cells. Requires a brief incubation of 15 mins at 37 degrees after which you can detect the mitochondrial superoxide using a plate reader.

Once everything is prepared for the assay (Krebs medium could be pre-made, aliquoted and stored at -20), it is a relatively easy experiment to perform. Whilst cells are in the plate reader, make sure to prepare everything for the protein assay (sufficient volume of Bradford reagent, etc.). Overall, it's a precise probe to use for the detection of mitochondrial superoxide.
