This study seeks to validate functional genomic mutations observed in human cancer cells resistant to drug therapies. This antibody was used in Western blotting of human cancer cell lysates.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Human cancer cell lysates

Primary Incubation

1:1000 in 5% non-fat milk in TBS-T, overnight, 4 degrees Celsius

Blocking Agent

5% non-fat milk in TBS-T, 1 hour, RT

Secondary Incubation

1:10,000 (anti-Ms) in 5% non-fat milk in TBS-T, 30 minutes, RT

Tertiary Incubation

N/A

Detection

Chemiluminescence HRP substrate

Results Summary

We observed bands at approximately 35 kDa, very near to the predicted size of Cyclin D1 (36 kDa). There are also very large non-specific bands near the top of the gels - these may be an artifact of the electrophoresis procedure and unrelated to the specificity of the antibody.

Summary

The Good

Recognizes band corresponding to the correct predicted size of Cyclin D1

The Bad

Observed non-specific bands

The Bottom Line

Overall, this is a decent antibody that I would purchase and use again.
