Molecular cloning
PCR product
PCR amplify insert with 15bp of homology on each end. Mix PCR product with cold fusion enzyme and plasmid. Transform into bacteria.
Follow the kit recommendations for insert to plasmid molar ratio.
I usually get tons of colonies, the majority of which are the right product.
