This is basically a glorified blender that dissociates any tissue digested with collagenase, to extract infiltrating cells. It can be used for tumors, pancreases, kidneys, heart etc. Some solid tissues are difficult to dissociate by manual agitation, and this is where it comes handy. Plus, who wants to hurt their hands when you have a million tissues to process? We actually performed a yield test and found that tumors dissociated with this instrument gave 5 times higher numbers of infiltrating leukocytes than manually agitated samples.

Protocol Overview

Chop the tumors using scalpels and incubate them with collagenase for 2 hours. After that, transfer the homogenate into C tubes (to be bought separately) and load them on the dissociator and run the tumor program. After 2 minutes, your samples are ready for ficoll separation.

Use 1 tube per organ
