We had the C1000 thermal cycler and upgraded it to the CFX90 real-time system to allow for performing of real-time PCR for quantitative gene analysis. It has a touch screen, which allows smooth programming & running; comes with a USB port for exporting data and an awesome software that will perform the data analysis for you. You just import data files into it and the software will calculate fold changes etc. I completely recommend this equipment to every lab as it is very reasonably priced.
Real-time PCR analysis
cDNA converted from RNA
Prepare a cocktail of your cDNA with SYBR green Master mix (2X), primers of interest and add water. Add the cocktail to a PCR tube or plate and run on the machine with the standard real-time program.
Normalize all your samples to equal amounts of RNA and not cDNA.
The machine shows consistent results for housekeeping genes, which makes the data trustworthy. All my triplicates have similar signal, suggesting that there is little or no technical variation.
