Fluorescence microplate readers are multimode spectrophotometers with the capability to detect and measure fluorescence emitted by analytes in a multi-well sample format. These instruments often feature additional detection modes in addition to fluorescence intensity, such as UV-vis absorbance, fluorescence polarization, and time-resolved fluorescence. Due to their ability to simultaneously measure multiple wells in a single run, fluorescence plate readers are useful for conducting various assays, such as quantitative assays, enzyme assays, and ELISAs. Considerations for readers include the measurement wavelength range, microwell plate capacity, throughput, detection methods, and a filter vs monochromator-based optical system. Instruments with a 96- or 384-well format will accommodate the most commonly used plates. However, for greater flexibility, some instruments can accommodate 6- to 1536-well plate formats.
