The activity of enzymes that catalyze polymerization of G-actin to F-actin was estimated using fluorescent labeled G-actin. Samples consisting of enzymes were prepared and mixed with fluorescent labeled G-actin in appropriate buffer. Theoretically, as G-actin polymerizes and make F-actin, there will be loss of fluorescence. So, kinetics of Fluorescent reading of sample mixture was read in Spectra max GEMINI XPS micro plate reader up to 5 minutes.

Protein sample were prepared and specific G-actin polymerization enzyme were immunoprecipited using specific antibodies. Precipitated enzyme was mixed with fluorescent labeled G-actin in suitable buffer and kinetics of fluorescence was measured.
Start machine at least 20 min before to get less noise.
Samples had shown a time dependent decrease of fluorescent readings indicating polymerization of G-actin to F-actin. Duplicate samples have showed almost similar readings with very low signal to noise ratio.
Can read plates up to 384 well formats. Good for kinetic and endpoint measurement. Good for temperature sensitive reactions as equipped with temperature sensor and controller.
