Endonuclease III, also called Nth due to the gene encoding it, is an iron-sulfur-containing enzyme. It is a DNA glycosylase that repairs oxidized pyrimidines. It also possesses apurinic/apyrimidinic (AP) lyase activity. The N-glycosylase activity releases damaged pyrimidines from double-stranded DNA, which results in an AP site. It then cleaves 3´ to the AP site leaving a 5´ phosphate and a 3´ ring opened sugar. Cleavage of the DNA backbone is done through beta elimination. Endonuclease III can remove urea, 5, 6 dihydroxythymine, thymine glycol, 5-hydroxy-5 methylhydanton, uracil glycol, 6-hydroxy-5, 6-dihdrothimine and methyltartronylurea.
