Benzonase is a genetically engineered exonuclease, originally extracted from the enterobacterium Serratia marcescens. It is a homodimer in structure with a monomer molecular mass of 30 kDa and contains two stabilizing disulfide bonds. Also known as ‘Golden nuclease’, it can digest nucleic acids – both DNA and RNA – into 5'-monophosphate terminated oligonucleotides 2–5 bases in length. It has no sequence specificity but it is reported to exhibit a preference for G-C rich regions in double-stranded DNA. It can be used to remove fragments of nucleic acids from recombinant proteins, digest native or heat-denatured DNA and RNA, reduce viscosity caused by nucleic acids, prepare samples for electrophoresis and chromatography, and assist in the purification of viral vectors and vaccines.
