ELISpot Assays

ELIspot assays, or enzyme-linked immunospot assays, are used for the specific and quantitative measurement of cytokine secreting cells, generally in response to stimulation. In a standard ELIspot assay, cells are placed in a plate coated with specific antibodies to the cytokine of interest. Upon secretion, these cytokines are bound and immobilized to the antibodies. Once the cells are removed, the cytokines can be detected by the addition of new antibodies conjugated to labels, such as horseradish peroxidase or alkaline phosphatase. The addition of a substrate produces visible localized spots, which correspond to the amount of cytokine-secreting cells.
