ATPases catalyze the hydrolysis of a phosphate bond in adenosine triphosphate (ATP) to form adenosine diphosphate (ADP). As ATP hydrolysis yields inorganic phosphate (Pi), the amount of Pi generated is directly proportional to the activity of the target ATPase. ATPase assays generally measure the change in absorbance by colorimetric detection. ATPase assay kits can be used for testing the efficiency of drug uptake, enzyme kinetic studies, and studying the effects of inhibitors.
