Description
Cellecta's CRISPR Human Genome 87K Knockout Library is a single-module, pooled sgRNA library targeting approximately 19,382 protein-coding genes and 1,859 microRNAs with 4 sgRNAs per gene (~87,000 sgRNA constructs). In addition to gene-targeting guides, it includes 1,000 non-targeting controls, 400 intron-targeting controls, 400 intergenic sgRNAs, and 400 safe-harbor sgRNAs targeting AAVS1 and CLYBL. sgRNAs were designed with the Broad Institute GPP CRISPick tool and ranked with Cellecta's proprietary algorithm based on the criteria of DeWeirdt et al. 2022, using the HECG tracrRNA design (an improvement over HEAT). Supplied as 200 µg plasmid DNA in the pRSG162-U6-sg-UbiC-TagRFP-2A-Puro lentiviral vector (U6 promoter driving the sgRNA; UbiC promoter driving TagRFP-2A-puromycin resistance). Cas9 is not included. Suitable for genome-wide dropout viability and other pooled CRISPR knockout screens in human cells.
