Casein Kinase Inhibitors

Casein kinases are a family of serine/threonine-selective enzymes that can be broken up into two families, casein kinase 1 (CK1) and casein kinase 2 (CK2). CK1 is involved in the Wnt signaling pathway, shuttling of transcription factors, circadian rhythms, DNA repair, and DNA transcription. CK2 is involved in cell cycle control, DNA repair, and regulation of the circadian rhythm. Overactive casein kinase signaling has been seen in various cancer types and thus inhibitors to both CK1 and CK2 are of interest to pharmaceutical companies. While some of the currently available inhibitors are specific to a particular family, others act on both or on multiple types of kinases.
