WNT signaling is essential for embryonic development and adult tissue homeostasis, and its aberrant activation drives several cancers, most notably colorectal cancer. My lab studies the molecular mechanisms of oncogenic WNT signal transduction, with the goal of identifying therapeutic vulnerabilities in colorectal cancer. Dishevelled 3 (DVL3) is a cytoplasmic scaffold that transduces WNT signals from Frizzled receptors to the β-catenin destruction complex. I used the Cell Signaling Technology Dvl3 Antibody (Cat# 3218) to monitor DVL3 levels in HEK293T cells carrying AXIN1 base-editing variants.

Results Summary

I used this antibody to blot whole-cell lysates from a panel of nine HEK293T lines carrying base-edited AXIN1 variants, together with non-targeting controls, all treated with WNT3A-conditioned medium and R-spondin 3. Lysates were prepared in a Triton X-100 buffer, and 50 µg per lane was resolved on a 3–8% Tris-acetate gel and transferred to nitrocellulose. Membranes were blocked in 5% milk/TBS-T, probed with the Dvl3 antibody at 1:1000 overnight at 4 °C, then with HRP-conjugated donkey anti-rabbit IgG (1:10,000) and detected with standard ECL. The antibody gave a single strong band at approximately 80 kDa, within the expected range for DVL3, with consistent signal across all nine lysates and no additional bands on the blot. Note that we did not include a DVL3 knockout or knockdown control in this experiment. Because DVL1, DVL2, and DVL3 are closely related paralogs of similar molecular weight, a DVL3-null sample would have been the best way to confirm that the detected band is DVL3 rather than a cross-reacting paralog. Readers who need definitive specificity should include such a control.

Summary

The Good

The antibody detects a single strong band of the expected size for endogenous DVL3 in whole-cell lysates. At 1:1000 with standard ECL, the signal was strong enough that no high-sensitivity substrate was needed, and background was very low. The single-band pattern is useful when screening a panel of lysates, since it makes the readout unambiguous, though band identity was not confirmed genetically.
