Product Name:

CellTrace™ Violet Cell Proliferation Kit, for flow cytometry

Catalog Number:

C34557

Labelling adherent MDA-MB-231 breast cancer cells with CellTrace Violet for cell identification and tracking in NK cell–tumour coculture assays.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

MDA-MB-231 breast cancer cell line

Primary Incubation

Add 0.1 µM CTV working solution at a maximum density of 1 × 10^6 cells/ml. Incubate for 20 minutes at 37 °C, protected from light. Quench the reaction by adding 500 µL of complete DMEM medium and remove the solution from the wells.

Blocking Agent

DMEM + 10% FBS

Secondary Incubation

N/A

Tertiary Incubation

N/A

Detection

Spectral flow cytometer Cytek NL2000

Results Summary

The CellTrace Violet dye provided very clear and consistent labelling of adherent MDA-MB-231 cells, with strong fluorescence achieved at a concentration well below the manufacturer’s recommended concentration.

Summary

The Good

Provides very bright and clear staining of adherent MDA-MB-231 cells, with excellent fluorescence even at concentrations well below the manufacturer’s recommendation

The Bad

None

The Bottom Line

An excellent choice for labelling adherent MDA-MB-231 cells, providing bright and consistent fluorescence at very low concentrations, making it particularly useful for cell identification in coculture assays.
