I used the NanoOrange™ Protein Quantitation Kit to quantify residual protein following IVT mRNA production. The assay was easy to optimize, highly sensitive for low protein concentrations, and provided reproducible results using a low-range standard curve.

A low-range BSA standard curve prepared by serial dilution worked well for quantifying residual protein. During the heating step, incubate samples in PCR tubes or microcentrifuge tubes rather than directly in the microplate to minimize evaporation before transferring to the plate for fluorescence measurement.

Results Summary

The NanoOrange™ assay was used to quantify residual protein after IVT mRNA production. A low-range standard curve generated by serial dilution provided sensitive and reproducible quantification.
