I used DOTAP to formulate mRNA lipoplexes for in vitro transfection and mRNA delivery studies. The lipid consistently formed stable complexes with mRNA and produced strong protein expression in HEK293 cells while allowing efficient optimization of lipid-to-RNA ratios.

DOTAP dissolved readily and reproducibly formed mRNA complexes. We optimized several lipid-to-mRNA ratios to balance particle size, encapsulation efficiency, and transfection performance. Freshly prepared formulations consistently produced the best results.

Results Summary

DOTAP efficiently complexed mRNA and generated stable nanoparticles suitable for cellular delivery. Optimized formulations produced robust EGFP expression in HEK293 cells as measured by flow cytometry in the FITC channel. The lipid also demonstrated excellent reproducibility during formulation optimization, making it suitable for evaluating different mRNA doses and lipid compositions
