Company:

Invitrogen

Product Name:

Dynabeads™ M-270 Carboxylic Acid

Catalog Number:

14305D

I used these M-270 carboxylic acid Dynabeads to covalently couple my antibody for immunoprecipitation. Coupling was efficient and the beads gave low background with clean pulldowns.

Experimental Design and Results Summary

Application

Immunoprecipitation (covalent antibody coupling)

Starting Material

Cell lysate

Tips

Activate the carboxylic acid groups with EDC/NHS before coupling; include a low concentration of Tween-20 in wash buffers to minimize nonspecific binding.

Results Summary

Covalent coupling of antibody was efficient, yielding beads that produced clean target pulldowns with low nonspecific background.

Summary

The Good

Antibody is covalently fixed, so no antibody leaching into eluates—cleaner blots and reusable beads.

The Bad

Requires a carbodiimide activation step, adding time compared to ready-to-use Protein A/G beads.

The Bottom Line

Excellent choice when you need a covalently coupled, low-background IP, worth the extra coupling step.
