We utilize a SOX9 antibody to measure SOX9 expression in mouse brain tissue (different brain regions of interest) to detect astrocyte reactivity together with other markers of astrocyte reactivity. SOX9 is highly expressed in mature astrocytes and can serve as a potential marker associated with astrocyte reactivity when its expression is elevated.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Mouse brain tissue (40-μm-thick coronal sections)

Primary Incubation

1:1000 dilution, incubated overnight at 4 °C

Blocking Agent

0.01 M PBS, 0.3% Triton X-100, and 3% normal goat serum

Secondary Incubation

Goat anti-rabbit Alexa Fluor 568 (1:1000) for 2 hours at room temperature

Tertiary Incubation

N/A

Detection

Leica SP8 confocal microscope with photomultiplier tube (PMT) and hybrid detector (HyD) detectors

Results Summary

The SOX9 antibody helps identify whether astrocytes in the mouse models we are studying possess a reactive phenotype. SOX9 is a transcription factor and is localized in the nucleus. Together with other markers of potential astrocyte reactivity (GFAP, S100B), elevated SOX9 expression serves as a marker of inflammatory activation of astrocytes.

Summary

The Good

Reliable results, and a low concentration (1:1000) is needed, which is cost-effective.

The Bad

The antibody is pricey, but no other negative results were detected.

The Bottom Line

The antibody is great and works reliably. Highly recommended.
