In my research, I investigate how bacterial cells construct and modify their outer layer. I utilized a specific dye to label particular protein-SnapTag fusions, which I then imaged using STED super-resolution microscopy. The objective was to observe the locations of these protein fusions with high precision at various stages of the cell cycle. The dye enabled us to visualize these differences clearly.
I used the dye for live-cell labeling followed by STED super-resolution imaging.

The SnapTag fusion showed clear labeling at the cell surface. Using STED microscopy, the resolution of the signal was greatly improved. This allowed us to visualize certain patterns of this protein during various cell cycle stages and also after certain cell wall-acting antibiotics treatment.
