CD9, which is also known as p24 / tetraspanin-29 is often positive on mature B lymphocytes. However, it is upregulated in cancer, e.g., in B-lymphoblastic leukemia/lymphoma (B-LL). We use it to distinguish mature B cells from B-LL.

Experimental Design and Results Summary

Applications

Flow cytometry

Sample

Blood and/or bone marrow

Primary Incubation

15-20 minutes in the fridge

Blocking Agent

5% FBS

Secondary Incubation

None

Tertiary Incubation

None

Detection

Flow Cytometer equipped with Violet laser

Results Summary

The results on the B cells are dim, with unclear resolution from the negative population, and a smearing effect (see image); however, the B-LL cells tend to overexpress this marker, so we can still detect them (in conjunction with other antibodies of course).

Summary

The Good

Available on this channel (V450).

The Bad

Dim signal - suboptimal resolution.

The Bottom Line

The antibody is just about OK, and it causes little compensation issues. Look around for alternatives before you buy this one.
