Activation of endogenous retrotransposons frequently occurs in cancer cells and contributes to tumor genomic instability. To test whether inhibition of retrotranspositions has an anticancer effect, we used treatment with the nucleoside reverse transcriptase inhibitor (NRTI) stavudine in mouse cancer model MMTV-HER2/Neu that spontaneously develops breast cancer. Stavudine dramatically extended progression-free survival in mouse model following an initial complete response to chemotherapy (DMAG-17). The 17-DMAG treatment led to the activation of heat shock response.

Results Summary

We could show that DMAG-17 as HSP90 inhibitor treatment led to the activation of heat shock response. The MMTV-HER2/Neu mouse model appears ideal for further assessment of the effects of NRTI on spontaneous breast tumor development, tumor response to chemotherapy with DMAG-17, and on progression-free survival.
