PEG buffer solution was prepared and used to isolate extracellular vesicles from conditioned cell culture media. Extracellular vesicles were successfully characterized and used in downstream biological analyses.

Experimental Design and Results Summary

Application

Isolation of exosomes/EVs from conditioned cell culture media

Starting Material

Conditioned media from cell cultures

Tips

Final solution used was 8% PEG, 0.5M NaCl in DI H2O

Results Summary

Extracellular vesicles were successfully isolated from culture media using PEG and their size distribution was measure via nanoparticle tracking.

Summary

The Good

Cheap, simple to prepare, and almost as effective as very expensive precipitation kits at a fraction of the cost.

The Bad

Slightly lower yield, and purity than commercial kits.

The Bottom Line

For the price, preparing isolation buffers from PEG is much more efficient at large scales than commercially available kits and performs almost as well. Useful for preliminary studies of cell-secreted vesicles.
