Product Name:

QUANTI-Blue™ Solution

Catalog Number:

rep-qbs

My work requires using cell lines with stable reporters that secrete the SEAP (secreted embryonic alkaline phosphatase) protein. Quanti-Blue solution offers quick, sensitive, and easy detection of SEAP protein.

Experimental Design and Results Summary

Application

Detect secreted SEAP (secreted embryonic alkaline phosphatase) protein from reporter cell lines

Starting Material

Tips

The QB buffer can appears squishy/slushy if not fully thawed in 37ºC for up to 2 minutes. The QB solution should be freshly prepared for each use, and should be pre-warmed to 37ºC before assay. High concentration of FBS in culture media may interfere with background signals. If working with suspension cell line, make sure to spin the cells down (5-10min) to avoid adding cells into the QB solution.

Results Summary

Using the THP1-Dual reporter mammalian cell line that secretes SEAP protein upon activation of NF-kB pathways, QB solution helps detect different levels of NF-kB activation upon stimulation by different small molecules at different concentrations.

Summary

The Good

Quick, sensitive, easy, and keeps cells intact for subsequent experiments if needed.

The Bad

Can give high backgrounds. So, may be hard to detect low stimulation.

The Bottom Line

Give quick, easy, and sensitive detection of SEAP protein. Highly recommended for SEAP reporter cell lines.
