Functional characterisation of T cell. HUman PBMC were stimulated with PMA/onomycin for 5 hours in the presence of BFA. Cells were then surface stained with CD3/CD4/CD8, fixed, permeabilized and then stained with IL-4. FIxation snd permeabilization using eBioscience™ Foxp3 / Transcription Factor Staining Buffer Set. Antibody was diluted in 1X permeabilization buffer.

IL-4 detection is rather difficult in general. The antibody shows very dim, yet specific, staining in PMA-Ionomycin stimulated lymphocyte. 1:200 is the sweet spot and could separate positive and negative population. Higher concentration increase the background. Lower concentration is not recommended.
