Phospho-Histone H2A.X (H2A histone proteins) antibody was used in rat peripheral blood mononuclear cells (PBMCs) samples.

Experimental Design and Results Summary

Applications

Immunohistochemistry

Sample

Rat PBMCs

Primary Incubation

1:200

Blocking Agent

Normal Goat Serum diluted in 0.1% BSA

Secondary Incubation

biotinylated secondary antibody (1:200)

Tertiary Incubation

Fluorescein avidin‐DCS (1:500)

Detection

Confocal - fluorescence

Results Summary

pH2AX antibody was able to discriminate foci in rat PBMCs. We noticed that the fixation of the sample affects the staining. Prior to staining we incubated the samples with 1% Triton X-100.Optimization was required for different tissues.Image: pH2AX (green), DAPI (nuclei).

Summary

The Good

Good dilution.

The Bad

In some tissues it is difficult to detect.

The Bottom Line

Sometimes foci are difficult to detect, although a "blur" of staining may be found in the nuclei of the cells.
