The Mitotracker Red CMXRos Dye from Invitrogen is a very high-yield dye for staining of Mitochondria in vitro. The Dye is quite stable and bright and can be used in fixed or viable cells without problems.

Tips

Start at a very low concentration (100 nM is usually enough), make sure to wash out unbound dye with fresh medium, possibly fix cells afterwards.

Results Summary

The dye is even at low concentrations very bright (100 nM, 30 min incubation). In fixed cells the dye remains stable and is perfectly suitable for IF stainings (excitation in red channel e.g. 552 nm laser). However, since its so bright and high-yield it is sometimes not 100 % specific. Optimization with different concentrations might be necessary. Quality of results depends on the sample.

If you ever wanted to stain mitochondria in a quick and easy manner I totally recommend the Mitotracker Red staining. In contrast to Mitotracker green it can be combined with fixation and usually gives you bright signals.
