To study mitochondrial proteins and their activity, I isolated mitochondria from cells. To check the purity of mitochondria and the efficiency of the mitochondria preparation protocol, I used the VDAC antibody as a control.
Western blot
Neuro2a cells
1:10000 in 1% BSA, Overnight at 4°C
5% BSA in Tris-buffered Saline Tween-20
1:10000 HRP-conjugated anti-Rabbit IgG, 1 hour at RT
ECL detection in documentation system (Vilber Lourmat).
The antibody gives a clean band for VDAC in total cell fraction and mitochondrial fraction. It is absent from the cytosolic fraction.
Aliquot and freeze for longer storage.
Works in higher dilutions, and gives a good and clean signal.
