In order to clone bacteria genes, I must extract genomic DNA (gDNA) using this kit, which is more efficient than the traditional cetyltrimethylammonium bromide (CTAB) method.
Genomic DNA isolation
Overnight grown bacterial culture
1 milliliter of over grown culture is sufficient. Do not take more culture for the prep.
I isolated many gDNAs from my gene knockout bacterial library and verified the strains by PCR and DNA sequencing. This could be possible only because of pure yields.
