I used the MTT assay as a simple, rapid, and reliable indicator of the cell viability of cultured cells under different treatment conditions.

Use negative and positive controls.
Cells were plated in 96-well culture plate. After treatment, MTT solution (0.5 mg/ml in growth medium) was added followed by incubation for further 3 h under normal growth conditions. Medium was removed and formazan crystals were dissolved in DMSO. Absorbance was measured at 570 nm using a plate reader.
