Product Name:

SuperSignal™ West Dura Extended Duration Substrate

Catalog Number:

34075

The SuperSignal™ West Dura Extended Duration Substrate is a very sensitive Chemiluminescence Substrate for detection of HRP-coupled antibodies e.g. in Western Blot. We use it for all our Western Blot membranes and are very satisfied with brightness and life-time of the substrate.

Experimental Design and Results Summary

Application

Western Blot

Starting Material

Nitrocellulose membrane loaded with primary/secondary antibodies for Chemiluminescence detection (HRP-coupled)

Tips

We re-use the substrate multiple times, before replacing it, it is working up to 24 h after preparation.

Results Summary

We use approx. 3 ml substrate per membrane (in 50 ml tube) and incubate for 5 min at RT (rolling) before imaging. Usually detection times are between a couple of seconds and several minutes depending on your antibodies. Otherwise signal brightness is much better than for other substrates we tested and the prepared solution is stable for up to 24 h in our hands.

Summary

The Good

Brightness, life-time.

The Bad

Price, sometimes optimization needed (antibody-dependent).

The Bottom Line

I would recommend SuperSignal™ West Dura Extended Duration Substrate to anyone who needs a sensitive chemiluminescence substrate and is willing to pay a bit more for good signals. Bad antibodies or low protein amounts must no longer be a problem.
