I use this buffer to prepare DNA fragments for downstream cloning. Then I perform an agarose gel electrophoresis migration of the enzyme digestion products. I excise the DNA fragment of interest from the agarose gel and I purify it. It works very well.

Many restriction enzymes are active in this buffer, which is convenient to perform double digestion reactions. Moreover DNA modifying enzymes are also active in this buffer.
Gel electrophoresis migration of the digestion products confirms that all the DNA has been cut. I am able to perform cloning reactions with DNA digested in RCutSmart Buffer.
This buffer is compatible with a wide variety of enzymes.
